Abstract
A FLG knockout human induced pluripotent stem cell line was established using CRISPR/Cas12a genome editing.
- Loss of filaggrin function is linked to impaired skin barrier formation and inflammatory skin diseases.
- The edited iPSC line showed typical pluripotent stem cell characteristics and maintained the ability to differentiate into all three germ layers.
- Genomic stability was confirmed through karyotype and copy number variation analyses.
- The cell line was free of mycoplasma contamination.
- This model may facilitate research into filaggrin-associated skin biology and related diseases.
