Using gene editing to remove FLG in human stem cells to model skin barrier problems

Abstract

A FLG knockout human induced pluripotent stem cell line was established using CRISPR/Cas12a genome editing.

  • Loss of filaggrin function is linked to impaired skin barrier formation and inflammatory skin diseases.
  • The edited iPSC line showed typical pluripotent stem cell characteristics and maintained the ability to differentiate into all three germ layers.
  • Genomic stability was confirmed through karyotype and copy number variation analyses.
  • The cell line was free of mycoplasma contamination.
  • This model may facilitate research into filaggrin-associated skin biology and related diseases.
