Abstract
Fto expression was progressively reduced while global m6A levels increased during ischaemic injury.
- Fto was identified as a top negatively selected gene in neuronal cells under oxygen-glucose deprivation/reoxygenation.
- Neuron-specific knockout of Fto led to increased infarct size, neurological deficits, cognitive impairment, and neuronal cell death.
- RNA sequencing analysis indicated the activation of multiple signaling pathways associated with Fto deficiency.
- Atf3 was found to be hypermethylated and upregulated downstream of Fto loss, linking m6A dysregulation to higher neuronal apoptosis.
- Knockdown of Fto increased ATF3 mRNA methylation and expression, while Fto overexpression mitigated brain injury and improved neurological outcomes.
