Advanced precise engineering of immune cells to improve survival, movement, and cancer destruction

Updated

Abstract

Chimeric antigen receptor natural killer (CAR-NK) cells may offer a safer alternative to CAR-T cells due to their multi-antigen targeting capabilities.

  • Current limitations of CAR-NK cells include challenges associated with DNA double-strand breaks and p53-mediated fitness loss.
  • Next-generation editing technologies like base and prime editors can reduce genotoxic stress linked to conventional CRISPR methods.
  • Targeted transposon systems allow for programmable insertion strategies accommodating larger gene cassettes.
  • Synthetic and epigenetic circuits provide context-dependent control of gene expression, avoiding issues with tonic signaling.
  • Enhanced CAR-NK cell therapies could improve persistence, trafficking, and tumor eradication through advanced engineering techniques.
