Using CRISPR in living models to identify RPS19 as a target for immunotherapy in liver cancer

Updated

Abstract

Deletion of ribosomal protein S19 (RPS19) in hepatocellular carcinoma (HCC) cells mitigates the immunosuppressive tumor microenvironment.

  • RPS19 is identified as a key mediator of immune evasion in HCC.
  • Tumor-associated macrophages are the primary effector cells in the RPS19-mediated immune suppression.
  • Complement C5a receptor 1 (C5aR1) transmits signals from RPS19 to trigger macrophage-dependent immune suppression.
  • Targeted delivery of CRISPR-Cas9 using specially designed nanoparticles effectively reduces RPS19 expression in HCC cells.
  • This approach not only reverses immune suppression but also significantly suppresses HCC progression in mouse models.
