Gene editing screens under immune pressure identify CHD1 and MAP3K7 as factors linked to cancer immunotherapy resistance

Updated

Abstract

Essence

CHD1 and MAP3K7 loss may make cancer cells more vulnerable to anti-tumor T cells and immune checkpoint blockade.

Evidence

This preclinical and translational study used whole-genome CRISPR-Cas9 screens in tumoroid-T cell co-cultures, cancer cell models, cytokine contexts, a syngeneic mouse melanoma model, and patient tumor-response correlations.

Caveat

The therapeutic and biomarker claims rest mainly on model systems and correlative patient tumor data rather than prospective clinical testing.
