Cell and gene activity changes from activating dopamine-making enzymes in cell models of Parkinson’s damage

Updated

Abstract

CRISPR activation of DDC increased its expression in SH-SY5Y cells at 24 hours (p = 0.0496) and 48 hours (p = 0.0064).

  • Targeting DDC with CRISPRa did not significantly affect cell viability in HEK293 cells.
  • 6-hydroxydopamine (6-OHDA) treatment significantly reduced cell viability in SH-SY5Y cells compared to untreated controls (p = 0.0129).
  • CRISPRa targeting of GCH1 did not lead to significant changes in expression levels.
  • Despite increased expression of DDC, no significant differences in viability were observed between CRISPRa-treated cells and the 6-OHDA-treated PD control group.
  • Further research is necessary to explore if transcriptional changes result in functional effects on dopamine production.
