Over 1,300 genetic interactions were identified using the CRISPRi-TnSeq method.

  • CRISPRi-TnSeq maps genetic interactions between essential and nonessential genes across many bacterial strains.
  • The method identified 1,334 genetic interactions, including 754 negative and 580 positive interactions.
  • Seventeen pleiotropic genes were identified, with some potentially acting as genetic capacitors that mitigate phenotypic changes.
  • Essential gene knockdown can be compensated by redirecting activity through nonessential genes in the same pathway.
  • A balance is necessary between Z-ring formation and peptidoglycan synthesis for successful cell division.
  • The control of c-di-AMP influences cell wall synthesis and turgor pressure, affecting bacterial integrity.
