Abstract

Single-stranded DNA 7 (ssDNA7) inhibits Cas9 activity across multiple tested spacer sequences and genomic loci.

  • Inhibitory effect is achieved without needing to redesign the inhibitor for different target sequences.
  • Carbohydrate conjugation enhances the stability of the oligonucleotide and its inhibitory activity in liver-derived and cancer cell models.
  • Amplicon sequencing confirms the inhibition of endogenous editing in various cell models and normal liver-derived organoids.
  • Cell-preferential inhibition is observed in matched cell comparisons.
