Optimized CRISPR/Cas12a system using split guide RNA for detecting microRNAs without amplification

Updated

Abstract

The system achieved limits of detection of 68 fM for miR-155 and 98 fM for miR-10b.

  • Distinct effects of spacer split position were observed on signal output, reaction kinetics, and background signal.
  • Optimization of DNA activator design, Cas12a/crRNA ratio, and reaction conditions improved assay performance.
  • The platform enabled sensitive detection of breast cancer-associated miRNAs while maintaining excellent specificity against non-target miRNAs.
  • Strong discrimination of mismatched sequences was demonstrated.
  • The assay supported dual-target screening within a single reaction and showed reliable performance in serum-spiked samples.
