Precise DNA editing of LPA gene in modified mice prevents large DNA losses

Updated

Abstract

Essence

Cytosine base editing lowered circulating apo(a) in LPA-transgenic mice while avoiding most large deletions seen with CRISPR-Cas9 cutting.

Evidence

Preclinical genome-editing experiments in LPA-transgenic mice showed durable circulating apo(a) lowering, and SMRT-seq detected no large deletions at one LPA target site and <4% large deletions across multiple sites.

Caveat

The work is limited to transgenic mice and genomic safety readouts, so it does not establish human efficacy or long-term clinical safety.
