Editing efficiency for potato improved from 26.8% to 76.7% using an optimized base editor.
- Cytosine base editors (CBEs) were developed for potato to enable precise genetic modifications.
- Two highly active cytidine deaminases were identified with distinct characteristics for diverse editing applications.
- The editing efficiency was notably enhanced by employing a specific promoter and enhancer, and by using chromatin-modulating peptides.
- Co-expressing the human RNA demethylase gene hFTO improved chromatin accessibility, further increasing editing efficiency.
- An efficiency of 85.7% was achieved for simultaneous editing of two target nucleotides related to herbicide resistance in potato.
