Improving DNA base editing in potatoes by combining multiple optimizations

Editing efficiency for potato improved from 26.8% to 76.7% using an optimized base editor.

  • Cytosine base editors (CBEs) were developed for potato to enable precise genetic modifications.
  • Two highly active cytidine deaminases were identified with distinct characteristics for diverse editing applications.
  • The editing efficiency was notably enhanced by employing a specific promoter and enhancer, and by using chromatin-modulating peptides.
  • Co-expressing the human RNA demethylase gene hFTO improved chromatin accessibility, further increasing editing efficiency.
  • An efficiency of 85.7% was achieved for simultaneous editing of two target nucleotides related to herbicide resistance in potato.
