Removing non-essential secretion-related genes improves protein production in Komagataella phaffii

Abstract

Strains of Komagataella phaffii with gene disruptions showed 2-3x enhancements in specific productivity of monoclonal antibodies.

  • A genome-wide knockout screen identified genes in K. phaffii that are less essential and related to protein secretion.
  • Disruption of selected genes linked to cell wall and vacuolar transport improved monoclonal antibody production.
  • No significant differences in growth or protein profiles were observed between modified strains and the parental strain.
  • The enhancements in productivity were consistent across various scales, culture formats, and processing operations.
  • Combining gene essentiality data with biological pathway knowledge can guide effective strain engineering for improved productivity.
