Abstract
A 17-amino acid sequence in the C-terminal region of DGAT2 is essential for its targeting to lipid droplets.
- DGAT2 is concentrated around cytosolic lipid droplets when lipid droplet formation is stimulated by fatty acids.
- Deletion of the identified 17-amino acid sequence prevents DGAT2 from relocating to lipid droplets, keeping it in the endoplasmic reticulum.
- Fusing the C-terminal sequence of DGAT2 to a fluorescent marker successfully directs the marker to lipid droplets.
- Replacing the corresponding region of MGAT2 with DGAT2's C-terminal sequence also facilitates its targeting to lipid droplets.
- DGAT2 in endoplasmic reticulum membranes is shown to be continuous with lipid droplets.
