Abstract
A manganese-enabled Cas12a platform allowed direct, amplification-free RNA detection, including ultrashort transcripts, and measured serum miR-21 in a lung cancer sample set.
This diagnostic platform study used structural and biochemical analyses across multiple Cas12a orthologues and tested clinical serum samples, reporting 60-fold higher trans-cleavage under Mn conditions than Mg conditions with femtomolar RNA sensitivity.
Clinical validation was limited to circulating miR-21 concordance and lung cancer discrimination, so broader diagnostic performance across diseases and real-world settings remains unproven.
