DNAzyme-activated guide RNA allows CRISPR-Cas12a to detect alkaline phosphatase and Staphylococcus aureus

Updated

Abstract

The platform achieved sensitive detection of alkaline phosphatase with a limit of detection (LOD) of 0.008 U/L and Staphylococcus aureus with a LOD of 29.76 CFU/mL.

  • A CRISPR-Cas12a biosensor was developed that does not require nucleic acid amplification.
  • The system utilizes a DNAzyme-activated engineered crRNA (e-crRNA) to enhance trans-cleavage activity.
  • Activation by Mn2+ allowed for selective detection and reduced non-specific activation from DNA hybridization.
  • A rational design of the split site improved the signal-to-noise ratio in detection.
  • The biosensor demonstrated capability for detecting both biochemical targets and bacterial cells.
