Donanemab treatment in early-stage Alzheimer's disease

Abstract

In a trial of 257 patients, donanemab produced a 3.20-point improvement in cognitive scores compared to placebo after 76 weeks.

  • The baseline cognitive scores were identical at 106 for both the donanemab and placebo groups.
  • After 76 weeks, the change in cognitive scores was -6.86 for donanemab compared to -10.06 for placebo.
  • Most secondary outcomes did not show significant differences between donanemab and placebo.
  • Donanemab was associated with a reduction in amyloid levels by 85.06 centiloids and a small increase in tau load.
  • Amyloid-related cerebral edema or effusions occurred with donanemab, but most cases were asymptomatic.
