Links Between Amyloid Reduction, Tau Changes, and Clinical Results After Donanemab Treatment

Abstract

The rate of donanemab-induced amyloid reduction at 24 weeks was moderately correlated with the amount of baseline amyloid (Spearman correlation coefficient r, -0.54; P < .001).

  • Baseline amyloid levels were directly associated with the magnitude of amyloid reduction.
  • A greater chance of achieving complete amyloid clearance was inversely associated with baseline amyloid levels.
  • Donanemab slowed tau accumulation in a region-dependent manner.
  • There was a significant association between percentage amyloid reduction and clinical change on the Alzheimer's Rating Scale, specifically in APOE ε4 carriers.
  • Modeling suggests amyloid may not reaccumulate to the 24.1-centiloid threshold for an average of 3.9 years after stopping treatment.
