Donanemab treatment linked to changes in blood markers in early Alzheimer's symptoms

Abstract

Plasma levels of phosphorylated tau217 and glial fibrillary acidic protein were significantly lower with donanemab treatment compared with placebo after 12 weeks.

  • A total of 272 participants, aged 60 to 85 years, were included in the trial.
  • Significant reductions in plasma pTau217 were observed (least square mean change difference vs placebo, -0.04; P = .002).
  • Glial fibrillary acidic protein levels also significantly decreased with donanemab treatment (least square mean change difference vs placebo, -0.04; P = .01).
  • No significant differences in plasma amyloid-β 42/40 and neurofilament light chain levels were found between treatment groups at the end of the study.
  • Changes in pTau217 and glial fibrillary acidic protein levels were significantly correlated with changes in amyloid levels following treatment.
