Dual-drug linked ionizable lipids for delivering genetic material and targeted treatment of brain tumors

Updated

Abstract

TVP significantly enhanced cerebral blood perfusion and exhibited superior brain accumulation compared to MC3-based LNPs.

  • A dual-functional ionizable lipid was developed by combining a neurovascular regulatory motif with the chemotherapeutic agent temozolomide.
  • TVP showed improved brain delivery and selective localization in glioma tissues.
  • The platform demonstrated intrinsic anti-glioma activity while effectively delivering nucleic acids.
  • Co-loading with therapeutic nucleic acids resulted in strong inhibition of glioma progression through combined targeting strategies.
  • TVP maintained favorable biosafety in both healthy and glioma-bearing mice.
