Ionizable Tetrahydropyrimidine Lipids for Effective mRNA Delivery

Abstract

A library of 26 ionizable lipids was synthesized in just 3 hours for enhanced mRNA delivery.

  • THP LNPs exhibited tunable particle sizes, favorable ζ-potentials, and high encapsulation efficiencies.
  • THP1 demonstrated the highest transfection efficiency after intramuscular administration, comparable to a conventional benchmark.
  • Optimization of THP1 with phospholipids significantly improved intramuscular mRNA delivery and sustained protein expression for up to 5 days.
  • THP1 LNPs successfully enabled intravenous delivery in a dose-dependent manner with minimal toxicity.
  • In a mouse model, THP1 LNPs were able to edit genes in specific liver tissues.
