Liver regeneration improved by delivering two genes using GalNAc-coated lipid particles to reduce MKK4 levels

Abstract

GalNAc-LNP-siMKK4 effectively promotes liver regeneration and reduces hepatocyte apoptosis.

  • Inhibition of the MKK4 protein is critical for liver cell growth and regeneration.
  • MKK4-siRNA (siMKK4) was developed for targeted knockdown of MKK4 to minimize off-target effects.
  • N-acetylgalactosamine (GalNAc)-modified lipid nanoparticles were utilized for targeted delivery of siMKK4 to liver cells.
  • GalNAc-LNP-siMKK4 demonstrated good stability, biocompatibility, and a strong ability to silence MKK4 expression.
  • The gene therapy system showed efficacy in promoting liver regeneration and improving liver function.
