Tracking and targeting liver cells with cholesterol-free lipid nanoparticles carrying DY635-modified lipids

Abstract

The targeted lipid nanoparticle (LNP) formulation demonstrated almost doubled mRNA expression in human HuH7 hepatocytes under optimized conditions.

  • The formulation strategy eliminated cholesterol, addressing the challenge of hepatic accumulation associated with conventional LNPs.
  • In vivo expression analysis showed altered expression profiles for the new formulation compared to traditional cholesterol-containing LNPs.
  • A targeting lipid was successfully conjugated and incorporated into the lipid nanoparticles at concentrations up to 1.1%.
  • Nanoparticle uptake occurred within 30 minutes, leading to effective gene expression across various cell lines.
  • Cellular uptake was efficiently tracked using fluorescent methods, confirming the successful introduction of the targeting moiety.
