Abstract

Condensates of the RNA-binding protein ATXN2 oscillate with the circadian cycle and enhance translation of circadian mRNAs.

  • Dynamic assembly of ATXN2 condensates is associated with increased robustness and phase coherence in a transcription-translation feedback loop.
  • Static condensates do not replicate the effects of dynamic condensate assembly on protein translation.
  • Condensates that oscillate at an appropriate phase shift can boost protein translation without reducing corresponding mRNA levels.
  • Two potential circuit designs that can achieve this timing include TTFL-regulated condensate transcription and a hybrid model involving an oscillatory condensate loop.
  • Residual 24-hour oscillations of ATXN2 condensates were observed even after knockdown of Per2, Bmal1, and Clock, supporting the hybrid model.
