The dysregulation of the AMPK-SREBP1-FASN (ASF) signaling axis is associated with abnormal fatty acid synthesis in metabolic dysfunction-associated steatotic liver disease ().

  • The ASF signaling axis plays a central role in linking metabolic disorders with immune inflammatory responses in MASLD.
  • Dysregulated ASF signaling and its lipotoxic products can impair liver cell function and disrupt communication between liver cells and immune cells.
  • This disruption may initiate a self-perpetuating cycle of metabolic and immune dysfunction that contributes to disease progression toward fibrosis and complications.
  • The specific impact of ASF dysregulation on the hepatic immune landscape is not yet fully understood.
  • Potential therapeutic strategies targeting the ASF axis could help restore balance in the metabolic-immune interactions involved in MASLD.
