Highly Sensitive Electrochemical Biosensor Using Multiple CRISPR/Cas12a Cutting Units

Updated

Abstract

A low limit of detection down to 2.1 aM is achieved for target miRNA using an enhanced CRISPR/Cas12a system.

  • The assembly of multiple CRISPR/Cas12a units via hybridization chain reaction (HCR) enhances detection sensitivity.
  • Improved cleavage efficiency is observed with the HCR/Cas12a system compared to a single Cas12a unit.
  • A tetrahedral DNA nanostructure on the electrode surface facilitates excellent reactivity and accessibility for cleavage.
  • The method demonstrates high specificity, storage stability, and accuracy for miRNA sensing in biological samples.
