Abstract

6PP3-PNP preferentially delivered mRNA to the spleen after systemic administration in mice.

  • Nine new hydrophobized polymers were synthesized for efficient mRNA delivery.
  • Three optimal polymeric nanoparticles were selected based on in vitro transfection screening.
  • Different nanoparticles showed varying organ tropism, with 6PP3-PNP targeting the spleen and others targeting the liver.
  • 6PP3-PNP demonstrated excellent biocompatibility in both in vitro and in vivo assessments.
  • The study highlights the structure-function relationship in hydrophobized polymers for mRNA delivery.
