Abstract

Event-driven pharmacology (EDP) may lead to sustained pharmacodynamic effects from transient binding events of plastogens.

  • Plastogens may produce long-lasting changes in neuroplasticity without subjective mental states or may cause such effects along with dissociative symptoms.
  • Neuropsychiatry evidence indicates that plastogens can activate synaptic machinery, enhancing the ability of synapses to respond to future stimuli.
  • Rapid-acting plastogens, such as ketamine and classical psychedelics, are proposed to operate within the EDP model.
  • Traditional drug development models focusing on stable receptor occupancy may not yield durable benefits and could increase side effects.
  • New methodologies for drug development and clinical dosing will be necessary to align with the distinct actions of plastogens.
