Disrupting daily light cycles changes daily patterns of clock and gene regulation in female mouse livers

Updated

Abstract

Disrupted light schedules led to a significant loss of rhythmic expression in 5 of 8 core clock genes in female SKH-1 mouse livers.

  • Impaired gene expression was observed throughout the 24-hour day in core clock and epigenetically regulated genes due to disrupted light schedules.
  • Dysregulated expression in core clock and epigenetic genes is linked to sleep-related diseases.
  • Rhythmic expression of core clock and epigenetic genes was confirmed under normal conditions in female mouse livers.
  • All active demethylases and Zfp57 lost rhythmicity following clock disruption.
  • De novo DNMTs, Dnmt3a and Dnmt3b, showed a slight damping of rhythmicity in mice with disrupted clocks.
