Epigallocatechin-3-Gallate may protect mice from fructose-related fatty liver by controlling cell death and recycling through SIRT5

Updated

Abstract

Epigallocatechin-3-gallate (EGCG) significantly mitigated hepatic lipid accumulation and inflammation in fructose-induced fatty liver disease models.

  • Ferroptosis and autophagy dysregulation are important factors in the progression of metabolic dysfunction-associated fatty liver disease (MAFLD).
  • EGCG restored SIRT5 expression, reduced iron overload and oxidative stress, and influenced key protein levels related to ferroptosis.
  • The treatment improved autophagy, indicated by changes in specific protein ratios and expression levels.
  • SIRT5 knockdown eliminated the protective effects of EGCG, suggesting these effects are dependent on SIRT5.
  • Molecular docking demonstrated a strong binding affinity between EGCG and SIRT5, supporting the proposed mechanism of action.
