Esketamine treatment significantly improved LPS-induced cognitive dysfunction and neurological injury.

  • Esketamine is associated with reduced cognitive deficits in a rat model of sepsis-associated encephalopathy.
  • The treatment inhibited the production of pro-inflammatory cytokines and decreased cell apoptosis in both animal and cell models.
  • Esketamine alleviated blood-brain barrier hyperpermeability and prevented endothelial leakage in response to LPS.
  • The protective effects of esketamine on blood-brain barrier integrity may involve the activation of the BDNF/TrkB pathway.
  • Inhibition of autophagy by esketamine was observed, and this effect was reversed by the addition of an autophagy inducer.
