Using engineered virus-like particles to deliver gene editing tools in pancreatic cancer cells

Updated

Abstract

Pancreatic ductal adenocarcinoma (PDAC) cell lines may be transfected using engineered virus-like particles (eVLPs) for gene editing.

  • PDAC cell lines have inherent resistance to traditional transfection methods.
  • CRISPR-Cas9 and prime editing can be delivered into PDAC cells using eVLPs.
  • The protocol allows for the packaging of CRISPR components with engineered guide RNAs.
  • Successful gene edits were demonstrated at the HEK3 locus and NQO1 exon 6.
  • This method can be adapted for targeting other genomic locations.
