How Gene Activity Patterns Link Exenatide Treatment to Aging Signs in Immune Cells in Colorectal Cancer

Updated

Abstract

The original exenatide-derived signature was associated with SenMayo scores in colorectal tumor macrophages after removing shared genes.

  • Reconstruction included 209,207 colorectal cells and 4316 tumor macrophages, with 17 patients eligible for analysis.
  • Removing CXCR2 and IL1B from both signatures preserved a strong positive association (Spearman ρ = 0.804; p = 0.00219).
  • The mean expression association exceeded most expression-matched random signatures, indicating a significant relationship (empirical p = 0.006).
  • Associations persisted after adjustment for inflammatory scores, but varied depending on the method of signature derivation.
  • The findings suggest that the association with inflammatory macrophages does not confirm drug-induced senescence.
