Abstract

Semaglutide is associated with statistically significant signals for depressive disorders in both the FDA and WHO databases.

  • Only semaglutide showed significant signals of disproportionate reporting for depressive disorders, with reporting odds ratios of 1.26 in FAERS and 1.38 in VigiBase.
  • Liraglutide and tirzepatide did not demonstrate any signals for depressive disorders.
  • Increased disproportionality for semaglutide was observed among female patients and reports from healthcare professionals.
  • The pattern of semaglutide-associated depression indicated an early failure pattern with no significant interactions identified with psychotropic medications.
  • Findings suggest that monitoring for safety should focus on specific drugs rather than assuming uniformity across the GLP-1 receptor agonist class.
