Modeling how cotadutide dose relates to nausea using a step-by-step prediction method

Updated

Abstract

The predicted nausea event rate at 600 μg of cotadutide is 25% with a biweekly titration scheme.

  • Gastrointestinal adverse effects, particularly nausea, are associated with GLP-1 receptor agonism.
  • Tolerance development through gradual dose increase may mitigate nausea incidence.
  • Three modeling approaches were used to analyze the exposure-nausea relationship, with two Markov models showing better performance than a proportional odds model.
  • Disease indications such as NASH and obesity, along with sex, were identified as influencing factors on nausea severity.
  • The biweekly titration with twofold dose escalation is suggested to be the most effective method to minimize nausea while reaching the therapeutic dose.
