Abstract

Lipid nanoparticles (LNPs) demonstrated improved targeting of pancreatic cancer cells through a dual functionalization strategy.

  • LNPs were designed to deliver either reporter DNA plasmid or mRNA specifically to pancreatic ductal adenocarcinoma (PDAC) cells.
  • The DNA plasmid utilized a chimeric promoter that selectively activates in PDAC cells.
  • Functionalization with folate and an anti-CD47 antibody targeted PDAC cells and inhibited immune evasion.
  • Both DNA and mRNA LNPs showed dose-dependent targeting of PDAC cells, with mRNA LNPs achieving higher transfection efficiency.
  • LNPs with anti-CD47 antibody enhanced phagocytosis and induced an inflammatory response in macrophages in mouse models.
