Blocking CD47 increases human immune cells' eating of lung cancer cells, especially after lowering other eating blockers using targeted RNA nanoparticles

Abstract

A single dose of siRNA-LNPs achieved significant downregulation of mRNA and protein for up to 7 days without reducing macrophage viability.

  • siRNA-LNPs effectively targeted multiple phagocytosis checkpoints in human macrophages.
  • Combination treatment with LILRB1-siRNA-LNPs and CD47 blockade enhanced phagocytosis of lung cancer cells.
  • Target mRNAs were downregulated in a dose-dependent manner, with repeated dosing leading to further reductions.
  • The approach did not compromise the viability of macrophages.
