Physical and protective changes in gut mucus caused by the absorption helper SNAC

Abstract

Exposure to sodium 8-[(2-hydroxybenzoyl)amino]octanoate (SNAC) increased the mucus retention force and viscosity of ex vivo porcine intestinal mucus (PIM).

  • SNAC altered the physical properties of PIM but not of ex vivo porcine gastric mucus (PGM).
  • The presence of SNAC decreased the permeation of cyclosporine A and ovalbumin through PIM.
  • In contrast, the viscosity and permeation of cyclosporine A and ovalbumin through PGM remained unaffected by SNAC.
  • Bulk permeation of fluorescein-isothiocyanate dextrans (FDs) through PIM was similar to that through biosimilar mucus (BM) in the absence of SNAC.
  • SNAC decreased the permeation of FD4 but increased the permeation of FD150 kDa through BM.
  • Nanoparticle confinement within BM was unaffected by SNAC, contrasting with observations in PIM.
