Stability in digestion and enzyme-blocking effects of new peptides from tilapia fish waste, with one peptide influencing hormone and transporter levels in gut cells

Updated

Abstract

Both LPCL and TPFLPDE peptides retained their DPP-IV inhibition capacity after gastrointestinal digestion.

  • Dipeptidyl-peptidase (DPP)-IV inhibition by specific peptides may aid in managing Type 2 diabetes by prolonging the activity of incretin hormones.
  • LPCL and TPFLPDE peptides were identified from tilapia viscera by-products and showed significant in vitro and in situ DPP-IV inhibition.
  • After gastrointestinal digestion, both peptides remained intact and maintained their ability to inhibit DPP-IV.
  • Post-DPP-IV pre-incubation resulted in decreased DPP-IV inhibition potency, suggesting a substrate-type inhibition mechanism.
  • LPCL was shown to modulate GLP-1 expression and increase PepT1 protein levels in STC-1 cells.
  • LPCL activated the PepT1 extracellular domain, enhancing receptor expression and interactions, as supported by molecular docking analysis.
