Gelatin Microribbon Hydrogels Help Stem Cells Build Bone Strongly over Many Levels of Stiffness

Abstract

Gelatin microribbon (μRB) scaffolds can be fabricated with stiffness ranging from 13-57 kPa.

  • Macroporous gelatin μRB scaffolds enhance mesenchymal stem cell (MSC)-based cartilage formation compared to conventional gelatin hydrogels.
  • Varying μRB stiffness did not significantly impact osteogenesis, as all stiffness groups supported upregulation of bone markers and collagen deposition.
  • Soft μRB scaffolds (13 kPa) lost structural integrity over time, while intermediate and stiff scaffolds maintained their shape and supported robust bone formation.
  • After 5 weeks of culture in osteogenic media, gelatin μRB-based scaffolds demonstrated a more than 10-fold increase in the compressive moduli of engineered bone.
  • Coating gelatin μRBs with hydroxyapatite nanoparticles accelerated the maturation of MSCs into osteoblasts and enhanced mineralization.
