Gene-edited autologous hematopoietic stem cell trials are concentrated in hemoglobinopathies, with one approved product and persistent editing-delivery and safety challenges.

Evidence

This registry-based review identified 44 interventional clinical trials of gene-edited autologous stem cell therapies, mostly for hemoglobinopathies (37) and fewer for immunodeficiencies (4).

Caveat

Because it maps registered trials rather than pooling outcomes, it cannot establish comparative efficacy and notes unresolved delivery, editing-efficiency, and off-target-editing challenges.
