Gene-edited hematopoietic stem cells show preclinical promise for leukemia and lymphoma but remain constrained by delivery, safety, and engraftment barriers.

Evidence

This PRISMA systematic review synthesized 19 preclinical and translational studies of CRISPR/Cas9, lentiviral, non-viral, base-editing, CAR-HSC, and safety-switch strategies.

Caveat

The evidence is not clinical efficacy evidence, and long-term engraftment, insertional mutagenesis, editing efficiency, delivery, manufacturing, and safety monitoring remain unresolved.
