A fast genetic modification and gene editing method using Agrobacterium rhizogenes for Liriodendron hybrid and its uses

Updated

Abstract

K599 Agrobacterium rhizogenes achieved a peak transformation efficiency of 60.38% in Liriodendron hybrids.

  • A streamlined transformation system was developed specifically for Liriodendron hybrids to address challenges with traditional methods.
  • Transformation efficiencies varied among different strains, with K599 being the most effective.
  • The optimized co-culture conditions involved 2 days and two-month-old seedlings, which yielded the highest transformation rates.
  • Transformation efficiencies for various Liriodendron hybrids ranged from 15.47% to 60.63%.
  • The system enabled successful CRISPR/Cas9-mediated gene editing, targeting the LhAQP1 gene.
  • LhAQP1-overexpression was linked to enhanced plant growth and drought tolerance, while gene editing increased dehydration sensitivity.
