Genetic differences and DNA changes linked to sleep-wake cycle disorders

Abstract

Of 871 screened articles, 38 studies were included in the systematic review on genetic and epigenetic factors associated with circadian rhythm sleep-wake disorders.

  • Delayed sleep-wake phase disorder was examined in 18 studies, while shift work disorder and non-24-h sleep-wake rhythm disorder were covered in 17 and 8 studies, respectively.
  • Genetic variants, particularly in core clock genes, were associated with increased susceptibility to circadian rhythm sleep-wake disorders.
  • DNA methylation changes in specific genes may influence circadian adaptation, especially in shift work disorder contexts.
  • Significant variability in study designs and definitions of disorders limits the ability to draw causal conclusions or compare findings across different disorders.
  • Only five findings met the review-defined evidentiary standards for genetic and epigenetic associations, indicating a need for more robust research.
