Essence

Essential-gene-coupled selection can enrich HDR-proficient cells and raise scarless CRISPR editing efficiency without drug or marker selection.

Evidence

Cell-platform experiments in HEK293 cells, primary epidermal keratinocytes, U2OS cells, and HeLa cells tested ssODN and plasmid donors, with HDR gains of 7-16-fold, 41-fold, and 6-34-fold across the stated contexts.

Caveat

The abstract shows cell-line and primary-cell editing efficiency, not in vivo delivery, off-target performance, or therapeutic safety.
