Genes that affect kidney filter cell survival under oxidative stress, found by genome-wide CRISPR screens

Updated

Abstract

The Adriamycin screen identified 819 depleted and 456 enriched genes in human podocytes.

  • Genome-wide CRISPR-Cas9 knockout screens were performed on differentiated human podocytes exposed to Adriamycin and hydrogen peroxide.
  • The hydrogen peroxide screen identified 277 depleted and 215 enriched genes.
  • Integrated analysis showed 27 genes were depleted in both injury models.
  • Candidate processes for podocyte survival include stress adaptation, chromatin regulation, mitochondrial metabolism, inflammatory signaling, and cytoskeletal organization.
  • These findings may help prioritize genes that regulate podocyte resilience to oxidative injury.
