Abstract

A total of 664 cis enhancer-gene pairs were identified through single-cell RNA sequencing.

  • The study mapped enhancer-gene pairs by using a multiplex approach with CRISPR/Cas9 perturbations.
  • Effects on gene expression were measured by analyzing 254,974 single-cell transcriptomes.
  • Of the identified pairs, 470 were classified as high-confidence connections.
  • These pairs showed enrichment for specific transcription factors and were located near their target genes.
  • The findings highlight a new framework for exploring the regulatory interactions in the human genome.
