GLP-1 Drug May Reduce Nicotine Reward by Changing Hunger and Digestive Hormone Signals in the Brain

Abstract

Nicotine administration (0.5 mg/kg) significantly increased conditioned place preference (CPP) scores in male ddy mice.

  • Nicotine is associated with increased expression of prepro-orexin and prepro-glucagon mRNA in the hypothalamus and nucleus tractus solitarius.
  • Liraglutide treatment (50 and 100 μg/kg) significantly reduced nicotine-induced CPP scores, indicating a decrease in addictive behavior.
  • Liraglutide downregulated the expression of prepro-orexin and prepro-glucagon, with the 100 μg/kg dose showing greater effectiveness in normalizing prepro-glucagon levels.
  • These findings suggest that liraglutide may mitigate nicotine reward by modulating neuropeptide pathways.
