Effectiveness of GLP-1-based drugs semaglutide, tirzepatide, and retatrutide in obesity caused by MC4R deficiency and their comparison

Abstract

Body weight reduction was 31.6% for tirzepatide in MC4R knockout mice after 21 days of treatment.

  • Semaglutide, tirzepatide, and retatrutide significantly reduced body weight in MC4R knockout mice by 19.7%, 31.6%, and 24.1%, respectively.
  • All three GLP-1 analogs suppressed both fat and lean mass in treated mice.
  • Improvements were observed in plasma insulin levels, cholesterol levels, and markers of liver damage, along with a reduction in liver hypertrophy.
  • GLP-1 analogs inhibited genes associated with fatty acid synthesis but did not significantly affect genes related to inflammation.
  • Energy expenditure decreased with GLP-1 analog treatment, with tirzepatide leading to a notable reduction in the respiratory quotient.
