GLP-2 may reduce postmenopausal bone loss by helping bone-building cells through a specific cell signaling pathway to lower FGF23 levels

Updated

Abstract

GLP-2 treatment significantly increased bone mineral density (BMD) by 18% in the femur and 22% in the lumbar spine of ovariectomized mice.

  • Ovariectomized mice exhibited lower femoral and lumbar BMD, reduced trabecular volume, and increased bone surface parameters compared to control.
  • GLP-2 treatment resulted in increased BMD, enhanced trabecular volume, and decreased adverse bone parameters.
  • GLP-2 upregulated Runx2 expression and downregulated FGF23 levels in bone and serum.
  • RNA sequencing indicated enrichment of the PI3K/AKT pathway among differentially expressed genes.
  • Inhibition of the PI3K/AKT pathway partially reversed the positive effects of GLP-2 on bone parameters and FGF23 levels.
