Effects of activating the glucagon-like peptide-1 receptor on mental health using genetic analysis

Abstract

Genetically predicted lower body mass index (BMI) via GLP-1R activation is associated with a 0.06 standard deviation improvement in mental well-being.

  • Lower BMI associated with GLP-1R activation correlates with a reduced risk of major depressive disorder (odds ratio 0.82).
  • There is a reduced risk of bipolar disorder linked to lower BMI via GLP-1R activation (odds ratio 0.61).
  • Suggestive evidence indicates that lower BMI through GLP-1R activation may also be connected to a decreased risk of substance use disorders.
  • The associations for mental health outcomes are stronger than those for lower BMI and lower glycated hemoglobin based on genome-wide variants.
  • Probabilities of colocalization indicate a significant relationship between BMI and mental health outcomes near the GLP1R gene.
